Triptorelin: A Synthetic Luteinizing Hormone Releasing Hormone Analog
Triptorelin is a synthetic analog of luteinizing hormone releasing hormone (LHRH), designed to mimic the actions of naturally occurring LHRH produced by the hypothalamus. Upon initial or intermittent administration, triptorelin stimulates the release of gonadotropins, including luteinizing hormone (LH) and follicle-stimulating hormone (FSH), from the anterior pituitary gland.
Long-term use of triptorelin is characterized by an initial phase of increased LH and FSH levels, followed by their suppression. In males, this leads to a temporary surge in testosterone concentrations. However, monthly administration of triptorelin in the treatment of prostatic carcinoma results in the suppression of LH and FSH secretion, leading to a decline in testosterone levels and pharmacologic castration within four weeks of the initial dose.
